【摘要】:Thistestmethodisfullyquantitativeanditalsoavoidsanylossofviableorganismsthroughwashoff.Thismakesitpossibletoproducestatisticallyvaliddatausingmanyfewertestandcontrolcarriersthanotherquantitativemethodsbasedonmostprobablenumbers(MPN).Thedesignofthecarriersmakesitpossibletoplaceintoeachapreciselymeasuredvolumeofthetestsuspension.Theuseofthethreadedstirbarsallowsforefficientrecoveryoftheinoculumevenafteritsexposureforseveralhourstostrongfixativessuchasglutaraldehyde.Themembranefiltrationstepallowsprocessingoftheentireeluatefromthetestcarriersandthereforethecaptureandsubsequentdetectionofevenlownumbersofviableorganismsthatmaybepresent.Thistestcanbeperformedwithorwithoutasoilloadtodeterminetheeffectofsuchloadingonmicrobicideperformance.Thesoilloaddevelopedforthistestisamixtureofthreetypesofproteins(highmolecularweightproteins,lowmolecularweightpeptides,andmucousmaterial)torepresentthebodysecretions,excretions,orotherextraneoussubstancesthatchemicalmicrobicidesmayencounterunderfieldconditions.Itissuitableforworkingwiththevarioustestorganismsincludedhere.Thecomponentsofthesoilloadarereadilyavailableandsubjecttomuchlessvariabilitythananimalsera.Sincethequalityoftapwatervariesconsiderablybothgeographicallyandtemporally,thistestmethodincorporatestheuseofwaterwithaspecifiedanddocumentedlevelofhardnesstoprepareuse-dilutionsoftestproducts.TheU.S.EnvironmentalProtectionAgencyrsquo;ScientificAdvisoryPanel(SAP)onGermicideTestMethodologyhasrecommendedtheuseofwaterwithastandardhardnessof400ppmasCaCO3.1.1Thistestmethodisdesignedforuseinproductdevelopmentandforthegenerationofproductpotencydata.Thistestmethodpermitstheloadingofeachcarrierwithaknownvolumeofthetestorganism.Theincorporationofcontrolscanalsodeterminetheinitialloadofcolonyformingunits(CFU)oforganismsonthetestcarriersandanylossinCFUafterthemandatorydryingoftheinoculum.1.2Thistestmethodisdesignedtohavesurvivorsandalsotobeusedwithaperformancestandard.Thesurvivingmicroorganismsoneachtestcarrierarecomparedtothemeanofnolessthanthreecontrolcarrierstodetermineiftheperformancestandardhasbeenmet.Toallowproperstatisticalevaluationofresults,thesizeofthetestinoculumshouldbesufficientlylargetotakeintoaccountboththeperformancestandardandtheexperimentalvariationintheresults.Forexample,ifanarbitraryperformancestandardof6-log10reductionintheviabilitytiterofthetestorganismisused,andaninoculumsizeof107CFU,thentheoreticallyamaximumoftensurvivorspercarrierispermitted;however,becauseofexperimentalvariability,theexacttargetmayneedtobehigherthan106CFU/carrier,thusfewersurvivorswouldbepermitted.1.3Thistestmethodshouldbeperformedbypersonswithtraininginmicrobiologyandinfacilitiesdesignedandequippedforworkwithinfectiousagentsattheappropriatebiosafetylevel(3).1.4Inthistestmethod,SIunitsareusedforallapplications,exceptfordistance,inwhichcaseinchesareusedandSIunitsfollow.1.5ItistheresponsibilityoftheinvestigatortodeterminewhetherGoodLaboratoryPracticeRegulations(GLPs)arerequiredandtofollowthemwhereappropriate(40CFR,Part160forEPAsubmissionsand21CFR,Part58forFDAsubmissions).1.6Thisstandarddoesnotpurporttoaddressallofthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyand......
